== Role in wound healing == Wounds to the skin will be repaired in part by the migration of keratinocytes to fill in the gap created by the wound. The first set of keratinocytes to participate in that repair come from the bulge region of the hair follicle and will only survive transiently. Within the healed epidermis they will be replaced by keratinocytes originating from the epidermis. At the opposite, epidermal keratinocytes, can contribute to de novo hair follicle formation during the healing of large wounds. Functional keratinocytes are needed for tympanic perforation healing. A 2026 phase I clinical trial involving five patients reported that allogeneic cultured keratinocyte sheets were feasible and caused no serious adverse events when used to treat deep second-degree burns.
== Coding and Homology == The very first nucleotide sequence of the gene encoding for the beta subunit of human chorionic gonadotropin (CGB) suggests that CGB evolved from a duplicate copy of the beta subunit of LH, another glycoprotein hormone with significant influence over pregnancy, expressed in the anterior pituitary gland. Subsequent nuclear mapping has found that humans possess six copies of the CGB gene, amazingly found together with the LHB gene on chromosome 19q13.33. Human CGB and LHB genes share an extremely high degree of similarity in their sequences, clocking in at 94% similarity. This nucleotide sequence is encompassed by 3 exons. Of the six genes that encode for CGB, four of them are functional. These include CGB, CGB5, CGB7, and CGB8. These four genes share a 97-99% DNA sequence similarity, and code for the biochemically functional beta subunit of hCG. Although CGB1 and CGB2 genes are similar in sequence to the other four genes previously mentioned (85%), they encode for a novel hypothetical protein that is 132 amino acids in length and does not share any homology to the functional CGB subunit. This particular result was caused by a DNA fragment insertion into the 5' untranslated region (UTR) of the CGB1 and CGB2 genes, giving way to a novel exon one and creating a single basepair open reading frame shift for exons two and three.

=== Pharmacodynamics === 4-HO-NiPT shows affinity for serotonin receptors, including for the serotonin 5-HT1D, 5-HT1E, 5-HT2A, 5-HT2B, 5-HT2C, 5-HT6, and 5-HT7 receptors (Ki = 229–2,461 nM). Conversely, it did not show affinity for the serotonin 5-HT1B or 5-HT5A receptors, whereas the serotonin 5-HT1A receptor was not reported. 4-HO-NiPT is a partial agonist of the serotonin 5-HT2A, 5-HT2B, and 5-HT2C receptors, with EC50Tooltip half-maximal effective concentration (EmaxTooltip maximal efficacy) values of 24 nM (88.4%), 188 nM (69.9%), and 963 nM (45.2%), respectively. == Mechanism of action == Melanotan II acts as a non-selective agonist of the melanocortin receptors MC1, MC3, MC4, and MC5. Melanotan II produces melanogenesis by activation of the MC1 receptor, whereas its clinically documented sexual effects are thought to be related to its ability to activate the MC4 receptor (though the MC3 is thought to also possibly be involved). Melanotan II is partly metabolised into Bremelanotide, a medication used to treat low sexual desire. Other effects of melanotan II, mostly regarded as adverse effects, include flushing, nausea, vomiting, stretching, yawning, and loss of appetite (the last via activation of MC4).

== Background and publication == A Scanner Darkly was one of the few Dick novels to gestate over a long period of time. By February 1973, in an effort to prove that the effects of his amphetamine usage were merely psychosomatic, the newly clean-and-sober author had already prepared a full outline. A first draft was in development by March. This labor was soon supplanted by a new family and the completion of Flow My Tears, The Policeman Said (left unfinished in 1970), which was finally released in 1974 and received the prestigious John W. Campbell Award. Additional preoccupations were the mystical experiences of early 1974 that eventually served as a basis for VALIS and the Exegesis journal; a screenplay for an unproduced film adaptation of 1969's Ubik; occasional lectures; and the expedited completion of the deferred Roger Zelazny collaboration Deus Irae in 1975. Because of its semi-autobiographical nature, some of A Scanner Darkly was torturous to write. Tessa Dick, Philip's wife at the time, once stated that she often found her husband weeping as the sun rose after a night-long writing session. Tessa has given interviews stating that "when he was with me, he wrote A Scanner Darkly [in] under two weeks. But we spent three years rewriting it" and that she was "pretty involved in his writing process [for A Scanner Darkly]". Tessa stated in a later interview that she "participated in the writing of A Scanner Darkly" and said that she "consider[s] [her]self the silent co-author".
